TRIONE Co., Ltd., founded in December 2022, is an innovative biotech venture developing first-in-class multi-target therapeutics based on the Brain-Muscle Axis. Building on original technology from Korea University, the company integrates an AI-driven drug design (AIDD) platform with medicinal-chemistry expertise to create single molecules that modulate multiple disease mechanisms at once. Our lead pipeline, Ph-30, is a first-in-class dual-target candidate that simultaneously targets Parkinson's disease progression and muscle wasting (sarcopenia). Ph-30 has completed preclinical proof-of-concept and is advancing into the IND-enabling stage. The company holds a strong IP portfolio, including PCT, U.S. (USPTO), and European (EPO) applications, positioning Ph-30 for global out-licensing to major pharma partners. By addressing large unmet needs in an aging society, TRIONE aims to become a world-class neurodegenerative-disease company.
Exhibit Description
Ph-30 is an orally available, small-molecule multi-target candidate for Parkinson's disease, based on a gamma-lactam (γ-lactam) derivative scaffold. A single molecule realizes a dual Brain-Muscle Axis mechanism: it inhibits brain MAO-B to protect dopaminergic neurons while blocking muscle Myostatin signaling to counteract muscle wasting. In preclinical studies, Ph-30 showed excellent blood-brain-barrier penetration (Brain/Plasma 1.8) and a wide safety margin with low cardiac liability (hERG IC50 > 100 μM). In the MPTP model, it significantly improved motor function on the rotarod versus reference drugs. Discovered via our AIDD platform, Ph-30 is produced with high optical purity (>98% ee), aiming to be a true disease-modifying therapy.
